I assume we are talking about stand-up and therefore I am not sure Simon Pegg can really be called the best. It has been said by many that Steve Martin is a stand-up legend. And then of course there is Richard Pryor.

The funniest female comedian I know of was the late Linda Smith.
It's all rather up in the air to be perfectly honest. I've not seen Larry David do stand up but I think Curb Your Enthusiasm is so funny that he couldn't not be on the list.
